Summary

This study investigated the effects of the peptide bioregulator prostamax on chromatin structure in human lymphocytes, using differential scanning calorimetry (DSC) and morphological analysis techniques. Prostamax is a regulatory peptide developed to act specifically on prostatic tissue, but its effects on DNA organization can be observed in other cell types.

The researchers treated human lymphocyte cultures with prostamax and analyzed changes in heterochromatin structure — the condensed and transcriptionally silent form of DNA. The calorimetry results revealed a redistribution of heat associated with chromatin phase transitions, indicating conformational changes in DNA organization.

The main findings included:

  • Partial relaxation of the chromatin fiber, evidenced by changes in thermographic profiles
  • Decondensation of heterochromatin regions, making them more accessible to the transcriptional machinery
  • Changes in nucleosomal organization consistent with gene activation

The significance of this study lies in demonstrating that small bioregulatory peptides can directly influence chromatin structure and, consequently, gene expression. This epigenetic mechanism is consistent with the peptide bioregulation theory which proposes that short peptides act as endogenous regulators of tissue-specific gene expression. The results provide a molecular basis for the therapeutic effects observed with prostamax in prostatic conditions, particularly benign prostatic hyperplasia in the elderly.
