Overview
Research peptide derived from the p53 tumor suppressor protein, designed to selectively bind to HDM-2 protein (human MDM-2) overexpressed on cancer cell membranes. Induces selective lysis of tumor cells without affecting normal cells. Used exclusively in oncology research contexts.
Scientific interest in PNC-27 centers on its mechanism of selective cytotoxicity against tumor cells: it binds to HDM-2 expressed on the membrane of cancer cells and induces lysis via pore formation, in theory sparing normal cells that do not express HDM-2 on the surface. In in vitro and in vivo models it has shown activity against several neoplastic lines.
PNC-27 is strictly a research peptide. It has no approval for clinical use in humans from any regulatory agency and should not be confused with approved oncology therapies. Its use is restricted to laboratories and preclinical studies; any use outside that context lacks support for efficacy or safety in humans.
It was designed from the tumor suppressor p53 activation domain (residues 12-26) fused to a cell-penetrating leader sequence (penetratin). Available data come essentially from preclinical studies; robust human clinical trials are still incipient.
